Statins need to be transported into cells by a protein made by the SLCO1B1 gene. A gene mutation in some people means less simvastatin is taken up by liver cells, increasing circulating blood levels. High doses can then result in muscle pain and weakness (statin myalgia/myopathy). Testing may reduce the risk of ADRs and drug discontinuation.8,20
